I am going to order the Earth Tone markers. I have seen a video on YouTube where the person used an Aqua Painter (which I have but have not used yet), SU Markers, and watercolor paper to make shading. I am wondering if this will work on regular card stock or only on watercolor paper.

While you can certainly watercolor on most "regular" card stock you will have 100% better results with watercolor paper IMHO.
My suggestion is to stop by Michaels, Hobby lobby, etc and pick up an inexpensive pad of cold press watercolor paper and play around with watercoloring. Once you do, you'll see the difference the paper will make.
